ABSTRAK
Pembangunan model biosensor DNA berasaskan pengikatan dan
interaksi kompleks logam sebagai penanda penghibridan DNA boleh
digunakan untuk diagnosis awal virus denggi melalui penghibridan DNA denggi
sebagai DNA sasaran. Kompleks nikel(II) salfen dengan rantai sisi
piperidina berjaya disintesis dan pencirian dijalankan dengan menggunakan
Spektroskopi Magnetik Nukleus (NMR), Spektroskopi inframerah (FTIR),
spektrometri jisim, dan kolorimetri pengimbasan pembezaan. Jenis perikatan dan
interaksi antara kompleks nikel(II) salfen dengan rantai sisi piperidina dan DNA denggi
dikaji dengan pentitratan DNA ultralembayung nampak. Berdasarkan
spektrum ultralembayung nampak, hipokromisme dan anjakan merah (batokromisme)
diperhatikan apabila kompleks nikel(II) salfen dengan rantai sisi piperidina
dititratkan dengan DNA denggi menunjukkan pembentukan
kompleks baru. Kompleks nikel(II) salfen dengan rantai sisi piperidina
berinteraksi dengan DNA denggi secara perikatan
interkalasi dengan pemalar perikatan, Kb= 4.576×105 M-1.
Penemuan ini sangat berharga dalam menjadikan kompleks nikel(II) salfen sebagai
penanda penghibridan bagi biosensor DNA optik untuk mengesan
kehadiran virus denggi.

ABSTRACT
Development
of DNA biosensor model based on interaction of metal
complexes as DNA hybridization marker can be used for dengue viruses
early diagnosis. Nickel(II) salphen complex with piperidine side chain were
successfully synthesized and characterized using Nucleus Magnetic Resonance
Spectroscopy (NMR), mass spectrometry, differential scanning
calorimetry and FTIR. The binding mode and
interactions of nickel(II) salphen complex with piperidine side chain and
dengue DNA were determined by UV-vis
titration. Based on ultra violet visible spectra, hypochromism and redshift
(bathochromism) were observed after upon tiration of the complex with dengue DNA which
suggested formation of new complex. Nickel(II) salphen complex with piperidine
side chain interact with dengue DNA by intercalation binding
mode with the binding constant, Kb= 4.576x105 M-1.
This new finding is so valuable for developing nickel(II) salphen complex as DNA hybridization
marker for DNA optical biosensor to determine dengue virus.
